I'm having exactly the same problem with a confermed bug found described on this thread:

I'm using Joomla 2.5.14 + Jomsocial 3.2.0.5 + JFBConnect 6.0.6 + JSPT 4.0.9
I'm following the thread waiting for a solution, and I thought maybe I can add details that can be usefull to help finding out what is wrong.
After a user register, (with Jomsocial Redirect plugin overrides on or off and registration link is not sent), if you try to actvate the new account manualy user will be able to login in the site, but the Jomsocial profile page url shortcut won't exist.
So, if you type
www.mysite.com/myuser
a System Message will say "User not found" (not a 404 error).
If you type
www.mysite.com/Jomsocial/go-to-profile
user will be able to see profile page.
I'm waiting desperately for this fix of the activation link and probably other people too, I don't know if those 2 things are connected, but hope it can be of any help.

2. As for redirection issue - it's caused by sh404SEF component. Disable it and issue should be gone.

I realized now that you did not fixed this problem, but used only a workaround.
You changed Joomla user activation to "admin" instead of "self".
This set up generated other issues with new users: the confirmed password is not being saved. It's conflicting also with other plugins I must have (Redirect to complete profile from ReadyBytes).
Also, this is not how the registration flow should be in my case. I'm not even receiving email notifications to approve new users (which seams to me another bug).
Therefore this is not a solution.
There's a real bug on it all and I hope you can investigate more in order to help me and other users.
For now I only can register users using facebook because of missing activation link.
Please help!

I have turned OFF plugins JFBConnect, Redirect to edit profile and JSPT
Email to the user is always missing activation link AND confermed password is not being saved.
If you activate the user manually user can make login and go to edit profile and receive the error message: "password and confirmed password are not the same". All tries to save the password are unusefull, it says "Your settings has being saved" but it DOES NOT save.
Also, when I click on Customize Profile in Jomsocial admin side menu I get a 500 error:
"String could not be parsed as XML"
Hope it could be a clue to investigate the problem further.

Well, this is not exactly a bug. As for now we have only TWO customers that reported this issue. So this is rather a site specific issue than a bug. None of us (support members) can replicate it nor developers can.
My workaround is working - user is receiving emails with activation link. And password works - I checked it too. Even now if you removed language constants I created.
Admin email will be not send and this is by design - not a bug.
Link is missing because you removed language constants I created....
And when I activated my account in backed, then tried log in I was prompt to complete my profile. So It works also...
In addition when I clicked "Cutomize profile" in backend I don't have any errors:

So I can't confirm any of your issues... and you rolled back changes I've made.

I really DID NOT rolled back any changes you made, excepted user to "self" approve on registration, as this is the set I need.
I also asked you please (in this thread) to tell me which were the changes you made, to not touch them acidentaly, but you did not replyied me.
I'm sorry you did your tests now, because I'm completely seting up every plugin again in order to find out a solution.
I will be unistalling everything in order to find out if it's really not Jomsocial's problem. Even if I'm the only one in world with this problem I really hope you to be able to help me.

After see your print screen I realized that I can access Customize Profiles if I use URL with no www, like: mydomain.com
I have reported this behavor to you in the past about older versions of Jomsocial.

I was thinking about this issue... and I noticed that in both cases customers uses JFBC plugins.
I disabled it on your site and activation set to "Self".
I received email with activation link:

Please confirm if work also for you.

It's not JFBConnect, when you tested it I had turned off JSPT.
I did other tests and confirm that.
I will be in touch now with ReadyBytes and post here any answers founded.

And I'd like to add an information in hope to help you track the bug: In my case I have created and enabled several custom profiles fields (including gender and birthday as you suggested) but system keeps sending COM_COMMUNITY_EMAIL_REGISTRATION instead of COM_COMMUNITY_EMAIL_REGISTRATION_REQUIRES_ACTIVATION in the exact same point of the logic.
To workaround it I did the following in com_comunity/controllers/register.php
